Description

Crystal Violet Test Card    

 

Principle

Crystal Violet rapid test kit is based on lateral flow immunochromatographic assay. The test device has a testing window. The testing window has an invisible T (test) zone and C (control) zone. When sample is applied into the sample hole on the device, the liquid will laterally flow on the surface of the test strip. If there is little Crystal Violet  in the sample, a visible T band will appear; T band is invisible if it has more Crystal Violet  in the sample. The C band should always appear after a sample is applied, indicating a valid result. By this means, the device can accurately indicate the presence of Crystal Violet in the water, urine, tissue ,fish,shrimp sample.

Solutions: Deionized water, Acetonitrile, N-hexane

 

 

Sample Preparation   

— water, urine, tissue ,fish,shrimp                                       

  1. Homogenize the sample.Take 4±05g in 15mL centrifuge tube, add 2.0mL NaCI,

shake for 2min properly.

  1. Add 150µL Sample Diluent, 4mL Acetonitrile, 2mL N-hexane, shake for 2min

properly.

  1. Centrifuge at 4000rpm for 5min at room temperature.
  2. Take 2mL middle layer to new tube, add 100µL Oxygenant,shake for 30s, dry by

nitrogen at 65°C .

  1. Add 0.3mL Sample Diluent, mix for 1min.
  2. Take 120µL lower layer for analysis.

 

Detect process

  1. Take out the cassette and microwell from the pack and place it horizontally.
  2. Return to room temperature (20-25℃), determine the cassette immediately.
  3. Move 120µL sample extraction to microwell,mix gently by shaking the plate

manually, place horizontally for 3-5min, become red solution.

  1. Move all red solution to card hole, react for 5-10 min and read result.

Result Judgement

  1. Positive:

The presence of “C” band, no matter T band. It means that Crystal Violet

in the sample extraction is higher than 2µg / kg .

 

  1. Negative:

Both clear“C” band and “T” band appears. It means that Crystal Violet

in the sample extractionis lower than 2µg / kg .

 

  1. Invalid:

No colored band appears in C zone, no matter whether T band appears

Note                                                            

  1. For best results, please strictly adhere to these instructions.
  2. All reagents must be at room temperature before running the assay.
  3. Do not remove test cassette from its pouch until immediately before use.
  4. Do not reuse the test kit.
  5. Do not use the test beyond its expiration date marked on the foil pouch.
  6. The components in this kit have been quality control tested as standard batch unit.
  7. Do not mix components from different lot numbers.

 

 

Storage and expiry date                                             

  1. Store at 4-30 °C for 12 months in the dark. Do not freeze.
