Nitrofuan(SEM) Rapid Test    

  

 

Principle

Nitrofuan(SEM)  rapid test kit is based on lateral flow immunochromatographic assay. The test device has a testing window. The testing window has an invisible T (test) zone and C (control) zone. When sample is applied into the sample hole on the device, the liquid will laterally flow on the surface of the test strip. If there is little Nitrofuan(SEM)  in the sample, a visible T band will appear; T band is invisible if it has more Nitrofuan(SEM)  in the sample. The C band should always appear after a sample is applied, indicating a valid result. By this means, the device can accurately indicate the presence of Nitrofuan(SEM) in the feed, urine, tissue sample.

Materials provided                                                 

Rapid Test Card  :        20units	HCl：        10mL
Derivatization Solution:    2mL	K2HPO4：     40mL
Sample Diluent ：        12mL	NaOH：      5mL

 

 

 

Materials required not provided                                        

Deionized water, Ethyl acetate, N-hexane

Centrifuge tube:  5mL, 10mL

Micropipettors:  20~200µL, 100~ 1000µL

Centrifuge, Balance(0.01g), Nitrogen evaporator

 

 

Sample Preparation                                             

1. Homogenize the sample.
2. Take 3±05g in 15mL centrifuge tube, add 3mL Deionized water,

0.3mL HCl, 200µL Derivatization Solution, shake for 1 min properly.

3. Incubate at 90°C water for 15min.
4. Add 2mL K₂HPO₄ and 0.2mL NaOH, mix 30s, then add 4mL Ethyl acetate,

shake for 1min completely.

5. Centrifuge at 4000rpm for 3min at room temperature.
6. Take 2mL supernatant to new tube, dry by nitrogen at 65°C .
7. Add 1mL N-hexane, add 0.6mL Sample Diluent,mix for 30s.
8. Centrifuge at 4000rpm for 5min at room temperature.
9. Take 120µL lower layer for analysis.

 

 

Sample Determination                                           

1. Take out the cassette and microwell from the pack and place it horizontally.
2. Return to room temperature (20-25℃), determine the cassette immediately.
3. Move 120µL sample extraction to microwell,mix gently by shaking the plate

manually, place horizontally for 3-5min, become red solution.

4. Move all red solution to card hole, react for 5-15 min and read result.

 

Result Judgement

 

1 .Positive: The presence of “C” band, no mat”ter “T

It means that Nitrofuan(SEM) in the sample extraction is higher than 0.5ppb.

2. Negative: Both clear“C” band and “T” band appears.

It means that Nitrofuan(SEM) in the sample extractionis lower than 0.5ppb .

3. Invalid: No colored band appears in C zone, no matter whether T band

appears.

 

Note                                                             

1. For best results, please strictly adhere to these instructions.
2. All reagents must be at room temperature before running the assay.
3. Do not remove test cassette from its pouch until immediately before use.
4. Do not reuse the test kit.
5. Do not use the test beyond its expiration date marked on the foil pouch.
6. The components in this kit have been quality control tested as standard batch unit.
7. Do not mix components from different lot numbers.

 

Storage and expiry date                                             

1. Store at 4-30 °C for 12 months in the dark. Do not freeze.
2. Expiry date is labelled on the package.
